Designed for oligonucleotide synthesis for diagnostic and therapeutic development
Thermo Scientific DNA phosphoramidites are built to meet the varying needs of oligonucleotide manufacturers for the development of therapeutic, diagnostic, and research applications. We offer TheraPure, standard, and fast deprotect phosphoramidites.
- TheraPure phosphoramidites undergo additional quality control release testing and are compared with our standard phosphoramidites, helping to ensure that impurities and residual solvents are controlled to the stringent levels required by our customers for their oligo therapeutics.
- Standard phosphoramidites support the quality and reliability suitable for routine oligonucleotide synthesis.
- Fast deprotect phosphoramidites allow the removal of base protecting groups under mild or accelerated conditions without the need for special reagents.
Our DNA phosphoramidites are supported by:
- Expert technical support—over 40 years of partnership, customization, and scale-up experience
- Secure and consistent supply—5x capacity expansion in 2022
- Service excellence—48 hours or less response to orders, >98% delivery to customer at promised date†
- Flexible packaging—custom and bulk packaging available upon request
- Quality—ISO 9001:2015–certified quality management system that has been refined in partnership with our customers since 1995
†May not be applicable in all regions.
Quality manufacturing and regulatory documentation
Processes are monitored from material procurement through final QC and product release for batch-to-batch reproducibility. Purity of each phosphoramidite lot is assessed by HPLC-MS, GC, 31P NMR, as well as Karl Fischer oven titration. Our rigorously controlled manufacturing processes help minimize levels of reactive P(III) impurities which could be incorporated into the oligonucleotide during synthesis and result in the accumulation of by-products.
TheraPure DNA Phosphoramidite highlights
- Raw materials and intermediates are always QC tested using analytical technologies (listed above)
- State-of-the-art purification methods utilized to obtain the highest amidite purity
- Overall, high purity and tight critical impurity level control yields excellent batch-to-batch reproducibility

Fast Deprotect DMF-dG, iPrPAC-dG, PAC-dA, Ac-dC, and iBu-dC phosphoramidites are ideal for applications requiring mild deprotection to preserve the integrity of oligonucleotides containing sensitive dyes, labels, or other modifications, which may be unstable with prolonged exposure to strong alkaline conditions. Fast Deprotect phosphoramidites permit the removal of base protecting groups under mild or accelerated conditions without the need for special reagents.
