Immunofluorescent analysis of Ras in HeLa Cells. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with a Pan Ras Monoclonal Antibody (Ras10) (Cat. No. MA1-012) at a dilution of 1:100 overnight at 4 C, washed with PBS and incubated with a Goat anti-Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, DyLight 488 (Cat. No. 35503). Ras staining (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Images were taken at 60X magnification.
GTP hydrolases (GTPases) are hydrolase enzymes capable of binding and cleaving GTP. Flow cytometry analysis of ADP-ribosylation factor in Hela cells compared to an isotype control (blue). Cells were harvested, adjusted to a concentration of 1-5x10^6 cells/mL, fixed with 2% paraformaldehyde, washed with PBS, and incubated with ARF1/ARF3/ARF5/ARF6 Monoclonal Antibody (1D9) (Cat. No. MA3-060) at a dilution of 1:80 for 60 min at room temperature. Cells were then blocked in a solution of 2% BSA-PBS for 30 min at room temperature, incubated for 40 min at room temperature in the dark using a DyLight 488-conjugated goat anti-mouse IgG (H+L) secondary antibody, and re-suspended in PBS for FACS analysis.
Western blot analysis of cdc42 was performed by loading 50 µg of various whole cell lysate onto a 4-20% Tris-HCl polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked with 5% Milk/TBST for at least 1 hour. Membranes were probed with a Cdc42 Polyclonal Antibody (Cat. No. PA1-092) at a dilution of 1:1,000 overnight at 4°C on a rocking platform. Membranes were then washed in TBS-0.1%Tween 20 and probed with a Goat anti-Rabbit IgG Secondary Antibody, HRP (Cat. No. 31460) at a dilution of 1:10,000 for at least one hour. Membranes were washed and chemiluminescent detection was performed using Super Signal West Pico (Cat. No. 34580).
