What is a plasmid midiprep?
A plasmid midiprep is a laboratory technique used to isolate and purify a moderate amount of plasmid DNA from bacterial cultures. It is a step between the smaller-scale miniprep and larger-scale maxiprep methods. The midiprep procedure typically involves similar steps to the miniprep, such as bacterial cell lysis, removal of cellular debris, and purification of the plasmid DNA. However, for the midiprep the overnight bacterial culture volume ranges from 25–100 mL and yields a higher quantity of plasmid DNA, usually in the range of 100–500 μg, making it suitable for applications that require a larger amount of pure plasmid DNA, such as transfection, cloning, or protein expression studies.
Deliver high-quality plasmid DNA from 25–100 mL of bacterial culture
When selecting a plasmid midiprep kit, scientists should prioritize high-quality plasmid DNA that is pure, intact, and free from contaminants. This helps ensure reliable results and reproducibility in downstream applications, fostering confidence in experimental results.
The GeneJET Plasmid Midiprep Kit enables large-scale isolation of high-quality plasmid DNA from recombinant E. coli cultures. With a spin column format and silica-based membrane technology, this kit offers convenience and efficiency, recovering up to 200 μg of plasmid DNA per prep. It is suitable for various molecular biology procedures, helping provide high yields and fast results in just 60 minutes plasmid DNA per prep.
The PureLink HiPure Plasmid Midiprep Kit isolates transfection-grade plasmid DNA from E. coli, yielding 100–350 μg of highly pure DNA. With no extra steps required to remove contaminants, this kit helps save time and labor. It is versatile, suitable for purifying various types and sizes of plasmid DNA, including BAC, bacmids, and ssM13 DNAs.
