NovaFluor Yellow 660 dye has been optimized for flow cytometers with spectral capabilities. Phiton technology provides highly stable fluorescence when stored at 4°C. NovaFluor Yellow 660 can be used to replace PE-tandems such as PE-Cy5 or PE-FireTM 640, with much less cross-excitation and fluorescent spillover. Replacement of PE-tandems with NovaFluor Yellow 660, which is primarily excited by the yellow-green laser, opens a detector, allowing for an additional marker to be detected off of the blue laser line using NovaFluor Blue 660.
Benefits of NovaFluor Yellow 660 dye include:
- Can be paired with medium to lowly expressed antigens
- Replacement for PE-Cy5 with reduced cross-laser excitation and fluorescent spillover
- PE-Cy5 can be replaced by NovaFluor Blue 660 and NovaFluor Yellow 660 for larger conventional flow panels
- Highly stable and compatible with all tested buffers
Use NovaFluor dyes with CellBlox Monocyte and Macrophage Blocking Buffer to block non-specific binding of NovaFluor labels, PE and APC tandems observed with macrophages and monocytes.

Experimental data using NovaFluor Yellow 660 dye products
Figure 2. Spectral signature of NovaFluor Yellow 660 dye. Normal human peripheral blood cells stained with anti-CD4 antibodies (clone SK3) conjugated to NovaFluor Yellow 660 dye were used for analysis. Data was acquired on a 5-laser Cytek Aurora system.
Figure 3. Staining of normal human peripheral blood cells with side scatter and unstained (left) or CD3 Monoclonal Antibody, NovaFluor Yellow 660 (Cat. No. H002T03Y04) (right). Data was acquired in the YG4 channel on a 5-laser Cytek Aurora and singlet cells were used for analysis.
