Accelerate and automate your method development using Thermo Scientific Vanquish Method Development HPLC and UHPLC systems. These systems can optimize separation conditions by testing different combinations of buffers on up to six HPLC columns. Vanquish Method Development HPLC and UHPLC systems use Thermo Scientific Chromeleon Chromatography Data System (CDS), which features downloadable eWorkflows to simplify automated column and buffer scouting while enabling secure and central data storage. Achieve fully automated method development with ChromSwordAuto Chromeleon Connect and ChromSword AutoRobust Chromeleon Connect. Chromeleon CDS is also compatible with the S-Matrix® Fusion QbD® software package.


Highlights

Automated column and eluent screening option

Screen up to 13 eluents by equipping your Vanquish Method Development HPLC and UHPLC system with a solvent selection valve for extensive, automated screening. The Thermo Scientific Viper Method Scouting Kit contains all the tubing and near-zero dead volume connections to screen four columns automatically without the need for any user interaction.

Universal and sensitive detection techniques

Detect all non- or semi-volatile analytes using a near-universal Thermo Scientific Charged Aerosol Detector. Additionally, gain more confidence in identification using the Thermo Scientific ISQ EC and EM single quadrupole mass detectors for peak identification and impurity profiling.

Automate, simplify and accelerate method development. The Vanquish Method Development HPLC and UHPLC systems are controlled by Chromeleon CDS and compatible with leading method development software packages, such as ChromSwordAuto Chromeleon Connect or Fusion QbD software.

The Chromeleon Data Vault ensures secure data storage and accessibility for users within the CDS infrastructure. With Vanquish Method Development HPLC and UHPLC systems, all data generated is safely backed up in Chromeleon CDS Data Vault, enabling easy data exchange throughout the organization.

Optimized for small and large molecules
When built using Vanquish Flex and Vanquish Horizon UHPLC components, these systems are biocompatible, enabling robust performance, even with high-salt gradients, while minimizing unwanted interactions between the analyte and system components. ChromSwordAuto Chromeleon Connect includes dedicated algorithms for more efficient separations of large biomolecules.

What are the stages of HPLC method development?

  1. Method scouting includes screening column and mobile phase conditions to choose the most promising combination for a successful HPLC separation. Traditionally, method scouting requires a significant amount of effort for manual column and mobile phase switching as well as instrument method creation. This process can be automated and streamlined using Viper fingertight fittings and solvent extension kits, and software packages such as ChromSwordAuto Chromeleon Connect and Fusion QbD.
  2. Method optimization, involving iterative testing of various separation conditions of the HPLC method, is performed to achieve the best possible resolution, speed, etc. Operators must run many samples and repeatedly make decisions based on results. This is the most time-consuming stage of method development and requires a great deal of user experience and knowledge. ChromSwordAuto Chromeleon Connect and Fusion QbD software significantly accelerates this step by rapidly analyzing separation data and automatically optimizing the HPLC method.
  3. Robustness testing is required to determine the impact of parameter variation on separation quality and is crucial for method validation. DOE and robustness testing can be automated using ChromSword AutoRobust Chromeleon Connect or Fusion QbD software.
  4. Method validation is the industry-specific process for determining whether a method is fit for purpose. Secure documentation of the method development process is necessary in a variety of industries. Chromeleon CDS ICH validation templates and documentation of all stages of the workflow can be stored in the secure Chromeleon Data Vault to ensure compliance.
