We offer versatile options for antibody purification of monoclonal antibodies including in vitro and in vivo ascites hybridoma production options for monoclonal antibodies. We also provide many service options for purifying the monoclonal antibodies that have been produced by these methods.
Following development of antibody-generating hybridomas, monoclonal antibodies can be produced from them by either (1) cell culture in T-flasks or bioreactors to yield antibody-containing supernatant or (2) mouse abdominal culture to yield antibody-containing ascites fluid. Each liter of cell culture typically yields approximately 10 to 30 mg of antibody. Each mouse abdominal culture yields 2 to 10 mg of antibody in 2 to 3 mL of neat ascites fluid.
Several options are available for monoclonal antibody purification from supernatants or ascites. Most customers prefer simple affinity purification with Protein A or Protein G resin, but we also can purify using anti-immunoglobulin antibodies or immobilized antigen.
Purification options for monoclonal antibodies
Monoclonal supernatants are purified under high salt conditions for stringent binding pending the antibody species and isotype. When the affinity of the antibody for an indicated capture resin is not known, small-scale pilots can be performed at additional costs.
- Standard purifications (included with in vitro production):
- Protein A/G or anti-immunoglobulin resin
- Protein A/G or anti-immunoglobulin resin with low endotoxin
Antibody labeling
We also provide integrated services for chemically labeling purified monoclonal antibodies to create conjugated antibodies for use in most assay systems. Let us know your needs and we can include this option when we provide a project quotation.
