MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases and contains N-terminal catalytic domain and C-terminal regulatory domain. MAP4K3 is upstream of the MEKK1 target and is stimulated by UV radiation and TNF-alpha and can lead to the activation of the JNK signaling pathway. MAP4K3 can directly activate PKC-T, during TCR signaling and people with systemic lupus erythematosus show considerable enhanced MAP4K3 expression and activation of PKC-T, and the kinase IKK in T cells. MAP4K3 can act as a pro-apoptotic kinase which orchestrates activation of BAX via the concerted posttranscriptional modulation of PUMA, BAD, and BIM. The kinase activity of the encoded protein can be stimulated by UV radiation and tumor necrosis factor-alpha. The protein specifically activates the c-Jun N-terminal kinase (JNK) signaling pathway. Evidence suggests that it functions upstream of mitogen-activated protein kinase kinase 1 (MEKK1). MAP4K3 previously was referred to as RAB8-interacting protein-like 1 (RAB8IPL1), but it has been renamed mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
