For maximum recovery please spin prior to use. Unless noted below, aliquots of the 5 µg, 10 µg and 20 µg sizes of kinase are not recommended as materials can be used in original packaging until exhausted. For larger sizes, the number of freeze/thaws may be reduced by preparing aliquots, aliquots below 20 µL are not recommended. Please never store a kinase diluted. If properly stored at -80C, this product is guaranteed for 6 months from date of purchase.
This recombinant fusion protein consists of full-length glycogen synthase kinase 3 alpha (GSK3A) and a glutathione S-transferase (GST) tag. It is validated in the LanthaScreen™ Eu Kinase Binding Assay. No special measures were taken to activate this kinase.

Easily Screen for Kinase Binding
The LanthaScreen™ Eu Kinase Binding Assay is a TR-FRET assay that lets you measure inhibitor binding to non-activated kinases, perform simple off-rate measurements and makes simple the study of type III (allosteric) inhibitors.
The LanthatScreen™ Eu Kinase Binding Assay is easy to optimize and run. The assay uses a simple mix-and-read format with a single one hour incubation and no development steps. Simultaneous binding of an Alexa Fluor™ conjugate or tracer” to a tagged kinase is detected by addition of a europium-labeled anti-tag antibody, in this case anti-GST. Simultaneous binding of the tracer and antibody to the kinase results in a high degree of fluorescence resonance energy transfer (FRET), while displacement of the tracer with a kinase inhibitor results in a loss of FRET (Fig 1).

Glycogen synthase kinase 3 alpha (GSK3A) is a multifunctional protein serine/threonine kinase implicated in the control of several regulatory proteins and transcription factors. (GSK3A Alias: DKFZp686D0638). Human GSTs can be divided into five main classes: alpha, mu, pi, theta, and zeta. The theta class includes GSTT1 and GSTT2. The GSTT1 and GSTT2 share 55% amino acid sequence identity and both of them were claimed to have an important role in human carcinogenesis. The GSTT1 gene is located approximately 50kb away from the GSTT2 gene. The GSTT1 and GSTT2 genes have a similar structure, being composed of five exons with identical exon/intron boundaries.
