For maximum recovery please spin prior to use. Unless noted below, aliquots of the 5 µg, 10 µg and 20 µg sizes of kinase are not recommended as materials can be used in original packaging until exhausted. For larger sizes, the number of freeze/thaws may be reduced by preparing aliquots, aliquots below 20 µL are not recommended. Please never store a kinase diluted. If properly stored at -80C, this product is guaranteed for 6 months from date of purchase.
This recombinant fusion protein consists of full-length hematopoietic cell kinase (HCK) and a glutathione S-transferase (GST) tag. It is validated in the LanthaScreen™ Eu Kinase Binding Assay. No special measures were taken to activate this kinase.

Easily Screen for Kinase Binding
The LanthaScreen™ Eu Kinase Binding Assay is a TR-FRET assay that lets you measure inhibitor binding to non-activated kinases, perform simple off-rate measurements, and makes simple the study of type III (allosteric) inhibitors.
The LanthaScreen™ Eu Kinase Binding Assay is easy to optimize and run. The assay uses a simple mix-and-read format with a single one hour incubation and no development steps. Binding of an Alexa Fluor™ conjugate or “tracer” to a tagged kinase is detected by addition of a europium-labeled anti-tag antibody, in this case anti-GST. Simultaneous binding of the tracer and antibody to the kinase results in a high degree of fluorescence resonance energy transfer (FRET), while displacement of the tracer with a kinase inhibitor results in a loss of FRET (Fig 1).

Hematopoietic cell kinase (HCK) is a SRC-related kinase expressed in lymphoid and hematopoietic cells and may play a role in some forms of leukemia. (HCK Alias: DKFZp686D0638). HCK is a protein-tyrosine kinase that is predominantly expressed in hemopoietic cell types. The encoded protein may help couple the Fc receptor to the activation of the respiratory burst. In addition, it may play a role in neutrophil migration and in the degranulation of neutrophils. Alternate translation initiation site usage, including a non-AUG (CUG) codon, results in the production of two different isoforms, that have different subcellular localization.
