For predictable high resolution separation of mono-, di-, and some oligosaccharides, use the Thermo Scientific™ Dionex™ CarboPac™ PA1 (1 mm) columns, when operating in Dual EGC mode on Thermo Scientific™ Dionex™ ICS-6000 HPIC™ systems equipped with Thermo Scientific™ Chromeleon™ CDS software. Dual EGC mode enables electrolytic generation of potassium methane sulfonate and potassium hydroxide gradients (KMSA/KOH) gradient eluent for carbohydrate analysis using a Dionex KOH EGC 400 KOH Eluent Generator Cartridge and a Dionex MSA EGC 400 MSA Eluent Generator Cartridge in series. Using the Dual EGC allows for an expanded set of samples for gradient separations of some oligosaccharides along with mono-, di-, and trisaccharide's using electrolytically generated potassium methane sulfonate and potassium hydroxide gradients with a combined concentration up to 200 mM. Many separations require only an isocratic separation. However, some groups of analytes will require a step or gradient elution. Retention of carbohydrates can be varied with eluent concentration, in some cases changing the elution order as the eluent concentration increases.
Combined with pulsed amperometric detection (PAD), this anion-exchange column provides fast, reproducible separations with no need for derivatization. The 1 mm (standard/microbore) Dionex CarboPac PA1, 1 mm columns for Dual EGC contain the same resin as the proven 4 mm and 2 mm Dionex CarboPac PA1 columns, which are used for non-Dual EGC carbohydrate analysis.
