Dynabeads MyOne CD45 Leukocyte Depletion is a highly efficient product designed for depletion of human CD45-positive cells from whole blood, buffy coat, or peripheral blood mononuclear cells (PBMC). After discarding the bead-bound cells, the product allows for utilization of the remaining untouched rare target cells, such as circulating tumor cells (CTCs), in downstream applications. The untouched CTCs obtained can be pelleted and lysed directly using the TaqMan Cells-to-CT Express Kit (Cat. No. A57988).
Dynabeads MyOne CD45 Leukocyte Depletion are uniform, superparamagnetic beads with a primary monoclonal mouse IgG2a antibody specific for all known isoforms of CD45, a membrane glycoprotein found on all human leukocytes. The product can be used to deplete CD45-positive cells easily and efficiently from viscous samples such as whole blood in approximately 30 minutes.
The Dynabeads MyOne CD45 Leukocyte Depletion magnetic beads process approximately 2.5 x 108 PBMCs per mL of beads.

Overview of the workflow for CTC enrichment
For CTC enrichment, Dynabeads MyOne CD45 Leukocyte Depletion is added to the sample under gentle tilting and rotation for 30 minutes to optimize binding to the leukocytes. The sample is then placed on a magnet for 5 minutes for separation of bead-bound leukocytes from the enriched CTCs. The supernatant containing the enriched CTCs is transferred to a new tube and the bead-bound leukocytes discarded. When enriching CTCs directly from whole blood, it is recommended to the red blood cells before the depletion (e.g., using Gibco ACK Lysing Buffer (Cat. No. A1049201)). Dynabeads MyOne CD45 Leukocyte Depletion is especially efficient in removing contaminating granulocytes from whole blood samples.
High throughput negative isolation of CTCs on KingFisher Purification Systems
Enrichment of circulating CTCs is a time-consuming process if working with many samples in parallel and can lead to increased manual handling errors. By utilizing the KingFisher Sample Purification Systems, you can achieve high-throughput recovery of untouched CTCs in approximately 50 minutes after loading the sample and reagents into the plates. The resulting CTCs are viable and suitable for culture, downstream molecular analysis, or proteomic analysis.
Alternative positive isolation strategies for CTCs
If it is preferred to positively isolate CTCs for molecular downstream applications such as RT-qPCR, we recommend using Dynabeads Epithelial Enrich (EpCAM). To positively isolate bead-free CTCs for immunological applications such as cell culture or flow cytometry, we recommend using Dynabeads CELLection Epithelial Enrich (EpCAM). If you are using your own biotinylated antibody, we recommend using Dynabeads MyOne Streptavidin T1.
