Getting Started
- Can anything be covalently labeled with a fluorophore, biotin, or linker?
- Which labeling chemistry should I use?
- My molecule has more than one reactive group on its surface. Can I control what sites are labeled?
- I labeled my protein with a fluorescent dye. Do I really need to do the ‘degree of labeling’ (DOL) determination?
- My molecule is insoluble in aqueous buffers. Can I still label my molecule?
Troubleshooting
- I labeled my protein with a fluorescent dye, but detect little or no fluorescence. Does this mean the labeling reaction was unsuccessful?
- I prefer not to use size exclusion chromatography to clean up my labeled protein after the labeling reaction. What other methods can be used for the post-reaction purification?
- I labeled my antibody with an amine-reactive dye, but it no longer binds to its antigen. What is causing this? Is there anything that can be done to prevent this from happening?
- My molecule precipitated during the labeling reaction. Why did this happen? What can be done to prevent this?
