Flow cytometry intrinsically facilitates the analysis of relative expression patterns in heterogeneous cell populations. Antibodies can be tested for staining in cell populations with known relative expression patterns. This representative image demonstrates the type of data that was used in the verification of specificity for this antibody. Legend: Balb/c bone marrow cells (top left) and splenocytes (top right) were surface stained with CD4 (clone RM4-5), CD45R (B220, clone RA3-6B2), and Gr-1 (clone RB6-8C5), followed by staining with 7-AAD. Viable bone marrow cells in the lymphoid (blue histogram) and myeloid (purple histogram) gates and viable splenocytes (orange histogram) were used for analysis. As expected based on known expression patterns, CD4 only stains a subset of splenocytes, CD45R stains subsets in both splenocytes and the bone marrow lymphoid gate but not the bone marrow myeloid gate, and Gr-1 only stains cells in the bone marrow myeloid gate.
