The Human Amyloid Precursor Protein (APP) ELISA quantitates Hu APP in human cerebrospinal fluid and cell lysates. The assay will exclusively recognize both natural and recombinant Hu APP.
Principle of the method
The Human APP solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Amyloid Precursor Protein (APP) or Amyloid beta precursor protein functions as a cell surface kinesin I membrane receptor, mediating the axonal transport of beta-secretase and presenilin 1. APP is important for neurite growth, neuronal adhesion and axonogenesis. APP is a 100-140 kDa transmembrane glycoprotein that exists as several isoforms resulting from alternative splicing. Proteolytic cleavage of APP by beta- and gamma-secretases results in the generation of beta amyloid, which is the primary component of senile plaques. Senile plaques are one of the major histopathologic features of Alzheimer's disease. Abnormal regulation and processing of APP also plays a role in Down's syndrome, early onset familial Alzheimer's disease, and cerebral hemorrhage.
