Description: The monoclonal antibody 8E11-SSEA5 recognizes human stage-specific embryonic antigen-5 (SSEA-5), an H type-1 glycan. SSEA-5 belongs to the SSEA family of glycoproteins and is highly expressed on pluripotent human stem cells including embryonic stem (ES) cells and induced pluripotent stem (iPS) cells. During embryonic development, SSEA-5 is expressed within the blastocyst on inner cell mass (ICM) cells that later give rise to ES cells. As ES and iPS cells differentiate, SSEA-5 expression is downregulated. The tumorigenic potential of contaminating stem cells in cell populations for transplantation is a hurdle to using this approach for disease therapy. The 8E11 antibody in combination with either CD9 and CD90 or CD50 and CD200 to sort populations has been shown to effectively separate teratoma-forming cells from stem cell-derived populations of differentiated cells.
Applications Reported: This 8E11-SSEA5 antibody has been reported for use in flow cytometric analysis, western blotting, and immunocytochemistry.
Applications Tested: This 8E11-SSEA5 antibody has been tested immunocytochemistry on unfixed human Ntera2 cells at less than or equal to 10 µg/mL. The 8E11-SSEA5 antibody has also been tested by flow cytometric analysis on NTERA-2 cells at less than or equal to 0.25 µg/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Stage-Specific Embryonic Antigen-5 (SSEA-5) is an oligosaccharide that can be part of glycosphingolipids and glycoproteins. It contains the motif Fuc1-2Gal-beta1-3GlcNAc-beta, which is characteristic of H-1 antigens. SSEA-5 is found in the inner cell mass of the human blastocyst during embryogenesis, on undifferentiated human embryonic stem cells and on the surface of induced pluripotent cells. SSEA-5 was found to proceed the loss of other known stem cell markers and the combination of SSEA-5 with either CD9 and CD90 or CD50 and CD200 was superior in identifying and eliminating rare undifferentiated human ES cells from differentiated cell populations than the use of other established stem cell markers.
