Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human GM-CSF. Anti-Human GM-CSF-specific antibody was purified by affinity chromatography employing an immobilized Human GM-CSF matrix.
Sandwich ELISA: To detect Human GM-CSF by sandwich ELISA (using 100 µL/well antibody solution) a concentration of 0.5-2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human GM-CSF (500-P33Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human GM-CSF.
Western Blot: To detect hGM-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 µg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant hGM-CSF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
The GM-CSF gene encodes for a cytokine that regulates the production, differentiation, and function of macrophages and granulocytes. The active form of the protein exists as a homodimer in the extracellular space. This gene is located in a cluster of related genes at the chromosome region 5q31, which has been associated with interstitial deletions in the 5q- syndrome and acute myelogenous leukemia. Other genes in the cluster include interleukins 4, 5, and 13. The gene is involved in promoting tissue inflammation. Elevated levels of cytokines, including the one produced by this gene, have been observed in SARS-CoV-2 infected patients with acute respiratory distress syndrome. Mice that lack this gene or its receptor have been shown to develop pulmonary alveolar proteinosis.
